【Animal Experiment】-Combined high-resolution MRI and micro-CT to evaluate the repairing effect of Taohong Siwu Decoction on rabbit femoral head necrosis

  OBJECTIVE: Combine high-resolution MRI and micro-CT technology to establish a rabbit femoral head hormonal necrosis model (SIONFH), and evaluate the repair effect of Taohong Siwu Decoction (THSWD) on SIONFH microscopic bone structure damage.

  Method: 25 New Zealand rabbits were randomly divided into 3 groups using a combination of endotoxin (LPS) and methylprednisolone (MPS). There are usually 5 groups, 10 model groups and 10 Chinese medicine groups (Taohong Siwu Decoction group). [Method] A hormonal femoral head necrosis model was prepared. Taohong Siwu Decoction suspension 0.3 g/kg was administered into the stomach in the Chinese medicine group, and the normal group and the model group were added with the same amount of ultrapure water. Be given. During the 8th week of the experiment, the femoral head was examined by high-resolution MRI using a 3.0T high-resolution magnetic resonance imaging instrument, and pathological examination was performed by hematoxylin-eosin staining and micro-CT of the femoral head.

  Results: 1) High-resolution MRI showed that the signal band at the femoral head in the model group was very low. The shape of the femoral head in the traditional Chinese herbal medicine group tends to be normal, the low-density area near the femoral skull epi is smaller, and the gray value of most bone tissues is normal. Compared with the normal group, the ROE value of the previous structural parameters between the model group and the normal group increased, and the ROE/NR ratio increased. Compared with the model group, the ROE value of the Chinese herbal medicine group decreased, and the ROE/NR ratio decreased. The difference is significant (P\u003c0.05). 2) Hematoxylin-eosin staining showed trabecular trabecular fracture, bone nucleus contraction, trabecular tear, bone marrow cell necrosis in the model group. The difference was statistically significant (P\u003c0.05). Traditional Chinese medicine shows that The osteogenic activity of the group was still relatively active, and the shape of cancellous bone was good. 3) Compared with the normal group, the micro CT, BV/TV, Conn.D., SMI, Tb.N, Tb.Th of the model group decreased, and BS/BV and Tb.Sp increased.. BS/BV of the traditional Chinese medicine group And Tb.Sp was significantly lower than the model group, Conn.D., SMI, Tb.N increased, the difference was statistically significant (P\u003c0.05). The three-dimensional reconstruction of OI showed that the model group had cancellous bone disease. You will see normal fractures and few bones. The bone quality of the herbal group was relatively improved, and the cancellous bone was repaired locally.

  Conclusion: Taohong Siwu Decoction can promote bone microstructure repair and biomechanical environment improvement in rabbit SIONFH model.
