【Animal experiment】-Deoxypurpletobacter inhibits the proliferation of colon cancer TH29 cells and induces apoptosis and its mechanism

  OBJECTIVE: To investigate the inducing effect of deoxylecithin on the proliferation and apoptosis of human colon cancer HT29 cells and its possible mechanism.

  Methods: HT29 cells were treated with different concentrations of deoxy violet and tetramethyl azo blue (MTT) colorimetric method to detect the inhibition rate of deoxy violin on HT29 cells; flow cytometry was used to detect cell cycle and WesternBlot method was used to detect apoptosis. Changes in the expression of death-related proteins Bax, Bcl-2 and Caspase9.

  Result: Deoxypurine has a significant inhibitory effect on the growth of HT29 cells. It increases the ratio of cells in the G0/G1 phase and decreases the ratio of cells in the S phase. Increase the rate of apoptosis, increase the expression of Bax and Caspase9 protein, and decrease the expression of Bcl-2 protein.

  Conclusion: Deoxypurine inhibits the proliferation of colon cancer HT29 cells and induces their apoptosis. The apoptosis mechanism may act on the internal pathway mediated by mitochondria.
