  OBJECTIVE: To study the expression level and significance of Maspin and IKKα in the mucosal tissues of the nasal sinuses of rats with fungal rhinosinusitis (FRS).

  Methods: 40 SD rats were established as fungal sinusitis models, and they were divided into nasal congestion group, FRS group, immunosuppressive group and aggressive FRS group according to the random number table method, with 10 rats in each group and 10 healthy rats Rats served as an empty control group; the control group was fed normally; the nasal congestion group was filled with hemostatic cotton and injected the same amount of 0.9% NaCl. Intraperitoneal and nasal injection; FRS group intraperitoneal injection of 0.9% NaCl injection; nasal injection of Aspergillus tobacco spore suspension; intranasal injection of 0.9% NaCl; intranasal injection of cyclophosphamide and Aspergillus fumigatus spore suspension to establish aggressive FRS , Established a rat model of aggressive FRS group. Enzyme-linked immunosorbent assay is used to detect the expression levels of serum interleukin-6 (IL-6) and tumor necrosis factor α (TNF-α). Immunohistochemical staining was used to detect the expression levels of Maspin and IKKα protein in the nasal cavity of each group of rats, and fluorescence quantitative PCR was used to detect the expression levels of Maspin mRNA and IKKαmRNA in the nasal cavity of each group of rats.   Results: The expression levels of serum IL-6 and TNF-α of rats in each group were significantly different (P0.05). From the results of immunohistochemical staining, the expression level of Maspin protein in the FRS group and the aggressive FRS group was significantly lower than that of the control group, nasal congestion and immunosuppression group, while the expression level of IKKα protein was significantly lower than that of the control group. Group, nasal congestion group, immunosuppression group (P\u003c0.05); Maspin protein expression in aggressive FRS group was significantly lower than FRS group, and IKKα protein expression was higher than FRS group (P\u003c0.05). The expression level of Maspin mRNA in the FRS group and the aggressive FRS group was significantly lower than that of the control group, nasal congestion and immunosuppression group. The expression level of IKKαmRNA was significantly higher than that of the control group, nasal congestion group and immunosuppressive group (P<u003c0.05); among them, the expression of maspin mRNA in the invasive FRS group was higher than that in the FRS group. The expression of IKKαmRNA was also significantly higher than that of the FRS group (P\u003c0.05).

  Conclusion: The down-regulation of maspin expression after activation of IKKα is the main cause of FRS, and this process may be one of the molecular mechanisms of invasive FRS.
