【Animal experiment】-Single intraosseous injection of simvastatin promotes normalization of breast cancer tumor blood vessels in mice

  Objective: To investigate the effect of a single intraosseous injection of simvastatin on mouse vasculature and breast cancer tumor function.

  Method: Establish an orthotopic model of mouse breast cancer and perform an intraosseous injection of simvastatin (50μg) or empty carrier. Alpha-SMA/CD31 immunofluorescence double staining was used to observe the pericyte/vascular endothelial cell range of tumor blood vessels. The vascular permeability of the tumor was observed by injecting Evans blue dye into the tail vein. Immunohistochemistry is used to detect changes in the expression level of HIF-1α in tumors.

  Result: Immunofluorescence double staining showed that compared with the control group, the simvastatin group (P\u003c0.05) increased the cell coverage of the tumor in the simvastatin group, with Evans blue in the tumor tissue. The content was significantly reduced (P\u003c0.05), indicating that the permeability of tumor blood vessels was reduced, and the tumor blood vessels became mature. Immunohistochemistry showed that the expression level of HIF-1α in tumors and breast cancer was reduced. The results showed that the lack of tumor tissue Oxygen is improved.

  Conclusion: A single intraosseous injection of simvastatin can increase the pericyte coverage of new blood vessels and promote the normalization of breast cancer tumor blood vessels.
