Objective: To construct the eukaryotic expression vector pEGFPN1/IL-37b of full-length mature IL-37b, and to detect its expression in RAW264.7 cells.
Method: Use the plasmid pUBC/IL-37b containing the IL-37b full-length coding region gene to construct a eukaryotic expression vector capable of expressing the full-length and mature form of IL-37b? The recombinant plasmid pEGFPN1/IL- was constructed. 37b was transfected into RAW264.7 cells, and the expression of full-length and mature IL-37b was detected by Western blot and confocal microscopy. Real-time PCR detects the type and inhibits IL-induced IL-6 expression.
"Result: The constructed pEGFPN1/IL-37b can express full-length and mature IL-37b in cells after transfection, and inhibit the expression of IL-6 induced by LPS.
Conclusion: Successfully constructed IL-37b full-length mature eukaryotic expression vector. This provides a solid foundation for further research on the anti-inflammatory effects and mechanisms of IL-37b.
