Methods: The acetic acid plus restraint stress method was used to establish post-inflammatory irritable bowel syndrome (PI-IBS) animal model; the rat colonic motility index (MI), the number of fecal particles excreted in 2 hours, and the expulsion time of glass beads were evaluated The exercise capacity of the rat colon; observe the formation of PI-IBS model rats and the therapeutic effect of Wuji Pills on it; use ELISA method to detect the calcitonin gene-related peptide (CGRP) in the brain and colon tissues of PI-IBS rats , Motilin (MTL), neuropeptide Y (NPY), substance P (SP), somatostatin (SS), vasoactive intestinal peptide (VIP), cholecystokinin (CCK) levels.
Result: The PI-IBS rat model was successfully established. The weight of model rats decreased; food intake decreased; stool volume increased; loose stools and amorphous soft stools appeared; voluntary exercise decreased; colonic MI increased significantly (P<0.05); the number of fecal pellets excreted by rats increased significantly (P <0.05); The discharge time of the glass beads is significantly shortened (P<0.05). After 7 days of treatment with Wuji Pills, the above symptoms can be significantly improved. Compared with the normal control group, the levels of CGRP, SS, and VIP in the brain tissue of PI-IBS rats increased significantly (P <0.05), and the concentration of NPY decreased significantly (P <0.05); while Wuji pills were administered Can significantly reduce the concentration level of CGRP, SS and VIP (P <0.05), significantly increase the concentration level of NPY (P <0.05). Compared with the normal control group, the levels of CCK, NPY, MTL, SS and VIP in the colon tissue of PI-IBS rats were significantly reduced (P <0.05); the administration of Wuji Pills can significantly increase the levels of CCK and VIP ( P <0.05).
Conclusion: Wuji pills can be used to treat IBS by regulating the levels of a variety of brain-gut peptides in the brain and colon tissues of IBS rats. These abnormally changed brain-gut peptides can be a potential biological diagnosis and treatment of IBS. index.
