Effect of Qingre Huatan Jiedu Recipe on Akt/JNK3/Caspase-3 and NF-κB signaling pathway in rats with stroke-related pneumonia

OBJECTIVE: To explore the effect of Qingre Huatan Jiedu Decoction on lung tissue of rats with stroke-related pneumonia, and its relationship with Akt/c-Jun N-terminal Kinase 3 (JNK3)/Caspase-3 (Caspase-3) ) And the relationship between the nuclear factor-κB (NF-κB) signaling pathway.

Methods: According to the random number table method, SD rats were divided into sham operation group, model group (cerebral ischemia reperfusion-lung injury rats), heat-resolving phlegm and detoxification prescription low (7mg/kg) and medium (14mg/kg) 、High-dose (28mg/kg) group, nimodipine group (200mg/kg), 24 rats in each group, hematoxylin-eosin staining (HE) was used to observe the histopathological changes of rat lungs, and to measure rat lungs Tissue wet and dry weight, arterial blood oxygen partial pressure; enzyme-linked immunoassay to determine lung tissue inflammatory factors and oxidation index levels, Western blotting (Westernblot), immunohistochemical detection of Akt, p-Akt, JNK3, p-JNK3, Caspase- 3、p65、p50 expression.

Results: Compared with the sham operation group, the dry weight/wet weight (W/D), neutrophil (PMN), malondialdehyde (MDA), and tumor necrosis of the model group Factor-α (Tumor necrosis factor-α, TNF-α), interleukin-1β (Interleukin-1β, IL-1β), lung tissue grade, lung tissue pathological score, lung tissue p-JNK3, Caspase-3, p65 、The positive expression rate of p50 protein and p-JNK3, p65, p50 protein increased, the arterial partial pressure of oxygen (PaO2), oxygenation index (OI), superoxide dismutase (Superoxide) dismutase, SOD), lung tissue p-Akt protein and protein positive expression rates are reduced; compared with the model group, the positive group, clearing heat and removing phlegm and detoxification group W/D, PMN, MDA, TNF-α, IL-1β, Lung tissue grade, lung histopathological score, lung tissue p-JNK3, Caspase-3, p65, p50 protein and p-JNK3, p65, p50 protein positive expression rate decreased, PaO2, OI, SOD, p-Akt protein and protein positive The expression rates were all increased, with a dose-dependent (P<0.05).

Conclusion: Qingre Huatan Jiedu Recipe can alleviate the pneumonia injury in rats with stroke-related pneumonia, and its mechanism may be related to the inhibition of JNK3/Caspase-3 and NF-κB signaling pathways and activation of Akt expression.
