[Animal Experiment]-Effect of Curcumin Pretreatment on Myocardial Oxidative Stress and Apoptosis in Rats with Heat Stroke in Dry Heat Environment

OBJECTIVE: To observe the effect of curcumin pretreatment on myocardial oxidative stress and cell apoptosis in heatstroke rats under dry heat, and to explore its possible mechanism

Methods: 50 male SD rats were randomly selected from 5 groups (n = 10): normal temperature control group (NC), dry heat control group (DHC), curcumin 50mg/kg pretreatment group (L-cur), curcumin 100mg /kg (M-cur), curcumin 200mg/kg (H-cur)? C? Did the DHC group receive normal saline gavage? Did the curcumin pretreatment group receive different concentrations of curcumin once a day for 7 consecutive days? On the 8th day, except the NC group, the other 4 groups of rats were transferred to an artificial laboratory in a special environment in the northwest. The ambient temperature is (41±0.5)°C, and the humidity is (10±1)%. After a 150-minute experiment, you will be in a state of heat stroke. After anesthesia, the materials are harvested and tested with an automatic biochemical detector. Changes in serum creatine kinase (CK) and lactate dehydrogenase (LDH) in rats, using TUNEL method to detect cardiomyocyte apoptosis and calculate the rate of cardiomyocyte apoptosis? Detection of myocardial superoxide dismutase (SOD)? Malondialdehyde (MAD) content and correlation analysis?

Result: Serum myocardial enzyme CK in dry heat control rats? LDH? Cardiomyocyte apoptosis index? MDA is significantly higher than the normal temperature control group (P <u\→ u0.01c0.01), turmeric CK? LDH? Cardiomyocyte apoptosis index? The MDA of the pretreatment group decreased with the increase of the drug concentration. The difference between the curcumin pretreatment group was significant (P<0.01); is the change trend of SOD opposite to that of MDA? Cardiomyocyte apoptosis and rat serum myocardial enzyme CK analysis showed that there was a positive correlation between LDH (correlation coefficient r = 0.30r = 0.30P<0.05); cardiomyocyte apoptosis and MDA were positively correlated (correlation coefficient r = 0.916, P<0.01), and there is a negative correlation between cardiomyocyte apoptosis and SOD (the coefficient is r = -0.86P<0.01)?

Conclusion: Curcumin pretreatment can affect dry heat environment, and heatstroke rats have a protective effect on myocardium. Can curcumin protect the heart muscle from oxidative stress? Do you want to reduce cell apoptosis?
