Objective: To study the effect of fluoxetine on the behavior and hippocampal neurogenesis in rats with post-stroke depression (PSD)?
Method: 32 adult male rats were randomly divided into sham operation group, stroke group, PSD group and fluoxetine. Did the Ting group use the middle cerebral artery embolization method to prepare a rat model of ischemic stroke, and a PSD rat model that combines chronic unpredictable mild stimulation with orphan methods? To assess the behavioral changes of rats at baseline, 7, 14, 21, 28, and 35 after surgery, they were weighed on the same day as the sucrose preference and the out-of-box test. A mandatory swimming test was added on the 35th day. At the end of the study, did you use immunohistochemistry to detect neuronal regeneration in the hippocampal dentate gyrus? Results: On the 28th and 35th days after the onset of stress, there were significant differences in the body weight, sucrose water consumption percentage, horizontal and vertical movement scores of the rats in the PSD group and the stroke group (P<0.05). There are significant differences in the oxetine group. Is the difference between the PSD groups also statistically significant (P<0.05)? Compared with the stroke group, does the immobility time of the PSD group rats in the forced swimming test increase significantly? Swimming and climbing time have been greatly reduced (P<0.01)? After fluoxetine fixation, is the fixation time of rats significantly shorter than that of the PSD group? Significant increase in swimming time (P<0.01)? Immunohistochemistry showed that compared with the stroke group (P<0.01) and the hippocampus of rats after fluoxetine intervention, the number of new neurons in the hippocampal dentate gyrus of the PSD group was significantly reduced. Compared with the PSD group, is there a significant increase in the number of new neurons in the dentate gyrus (P<0.01)?
Conclusion: Fluoxetine can effectively improve PSD rats and hippocampal depression. Does neuron regeneration participate in this process and play a key role?
