Why does TNBS modeling cause animal death?

      The following is the experience obtained by Zhonghong Boyuan in the experiment, and I am willing to discuss with you.

  1 TNBS 20-30mg/0.4ml/rat, in 50% ethanol for modeling (6w-old male SD rats), animal mortality is about 20-30%. After switching to 15-5mg/0.8ml/rat, in50% ethanol, no animals died.

  2 At the same dosage, reducing the concentration of ethanol can also reduce the inflammatory damage caused by TNBS. For example: 5mg TNBS in 50% ethanol can cause a small range of local ulcers and intestinal inflammation, but in 25% ethanol will not cause ulcers, only mild mucosal swelling.

  3 The depth of administration is usually 4cm or 8cm from the anus, but the administration distance seems to have little effect on the degree of acute inflammation.

  4 TNBS and ethanol need to be administered in a physiological saline solution, but the inflammation is more severe when administered in an aqueous solution, which may be affected by osmotic pressure.

  5 The catheter for administration must be smooth, because a non-smooth catheter may damage the mucous membrane, thereby aggravating the inflammation caused by TNBS.

   TNBS causes animal death mainly due to the following points:

  1 Intestinal perforation, abdominal infection. It may be improper operation or excessive local inflammation.

  2 Intestinal obstruction

  3 Intestinal adhesions and avascular necrosis of the intestine.
