[Animal modeling]-Intervention effect and mechanism of down-regulation of miR-192 on renal interstitial fibrosis in rats with unilateral ureteral obstruction

Objective: To investigate the effect of down-regulation of miR-192 expression on renal interstitial fibrosis in rats with unilateral urinary tract obstruction (UUO) and its mechanism

Method: Randomly simulated 45 SD rats were divided into the operation group and the model group antagomir-192 group. Except for the sham operation group, the other two groups of rats were ligated and the left urinary tract was ligated to obtain a rat renal interstitial fibrosis model. Rats in the antagomir-192 group were given Antagomir 192 tails 1-7-14 days after the operation. Rats in the sham operation group and the model group were given the same amount of normal saline via tail vein injection. On the 21st day, the rats were sacrificed by real-time fluorescent quantitative PCR (qRT-PCR) to detect changes in the expression of miR-192 in the occluded kidney tissue. HE staining was used to observe the serum urea nitrogen (BUN) and creatinine (Cr) content of rats, and observe the morphological and pathological changes of the obstructed kidney tissue. Masson staining was used to calculate the rate of collagen fiber deposition in obstructive renal matrix. Expression of markers related to mesenchymal transition (EMT) Cadoherin (E-Cadoherin), α-smooth muscle actin (α-SMA), Vimentin (Vimentin) and collagen type I (collagenI)

Result: Compared with the sham operation group, the expression level of miR-192 in the kidney tissue of rats in the model group and the expression of BUN in the serum? A model in which the Cr content, renal matrix injury score, and renal matrix collagen fiber deposition rate were significantly increased (P<001), and the expression of E-cadherin protein in renal tissue was significantly decreased (P<001). Compared with the group, α-SMA? The expression of vimentin and collagen I increased significantly (P<001). Compared with the model group, what is the expression level of miR-192 in the rat kidney tissue of the antagomir-192 group? Serum BUN? Cr content? Renal Interstitial Damage Score? The deposition rate of renal interstitial collagen fibers was significantly reduced (P), and the expression of E-cadherin protein in renal tissue was significantly increased (P).

Conclusion: The expression of miR-192 in the kidney tissue of rats with renal interstitial fibrosis increased, and the expression of miR-192 was down-regulated. The expression of vimentin and type I collagen was significantly reduced (P<001)001, which can inhibit renal interstitial fibrosis in UUO rats and play a protective role
