  【Principle of the experiment】Nitrosamine compounds are a kind of strong carcinogens, which can cause tumors in multiple organs in many animals. Some of these nitrosamines are strong inducers of esophageal cancer in animals, especially rats, and methylbenzylnitrosamine (NMBA) is the strongest carcinogen to rat esophagus among these compounds. NMBA has a special affinity for the esophagus, and the carcinogenesis it induces is limited to the esophagus. The specific mechanism is unclear.

   [Experimental materials] Animals: Wistar rats, both male and female, weighing 180g~220g. Drugs and reagents: methylpentyl nitrosamine, liquid paraffin.

   [Experimental method] Rats were injected subcutaneously with 1% methylpentylnitrosamine liquid paraffin solution 5mg/kg into the hind limbs once a week for 15 consecutive weeks.

   [Experimental results] Visually observe the changes in the esophageal mucosa from the pharynx to the stomach, with protrusions and papilloma formation, histological examination, the esophageal mucosa has proliferative lesions, precancerous lesions, and cancerous changes.

  【Matters needing attention】The model of induced esophageal squamous cell carcinoma is mainly made in rats, while mice are rarely used. There are also reports in the literature that Wistar rats are fed with sarcosine ethyl hydrochloride and sodium nitrite for modeling.

  【Evaluation】This model is often used to study the mechanism of some chemopreventive agents. Since the rat model of esophageal cancer induced by NMBA is histologically similar to human esophageal cancer, it is very valuable for studying the chemoprevention and carcinogenesis mechanism of esophageal cancer.

   Transplanted esophageal squamous cell carcinoma model

   [Experimental principle] Use human esophageal squamous cell carcinoma (HEC2 or Eca-109) tumor tissue to inoculate nude mice to establish an animal model of esophageal cancer.

  [Experimental materials] Animals: BALB/C nude mice, 6-8 weeks old, SPF grade, female. Drugs and reagents: human esophageal squamous epithelial carcinoma cell line Eca-109, human esophageal squamous cell carcinoma cell line HEC2.

   [Experimental method] A nude mouse with a diameter of about 1 cm was transplanted with human esophageal cancer (HEC2 or Eca-109) tumor tissue, cut into small pieces of about 1 mm3, and planted under the skin of the right axillary of the nude mouse.

   [Experimental results] The tumor quickly grows to a diameter of 3 to 4 mm3, which can then be used for drug screening experiments.

  【Notes】①The tumor tissue is cut into small pieces of about 1mm3 for experimental transplantation; ②The best tumor tissue is selected after 20 generations.
