Objective: To study the effect of acute exercise on the expression of miRNA-30a and autophagy-related genes in rat myocardium, and to explore the regulatory mechanism of miRNA-30a on myocardial autophagy activity in different recovery periods after exercise?
Method: SD rats are randomly divided into a group, in the quiet control group (Con), the 0h group (E0) after exercise, the 3h group (E3) after exercise, and the 12h group (E12) after exercise? Do you collect materials at different times after the acute exercise? Does real-time fluorescent quantitative polymerase chain reaction (PCR) detect the expression of ACE mRNA, miRNA-30a, Beclin-1 mRNA and LC3 mRNA in left ventricular muscle?
Result: The expressions of ACE mRNA and miRNA-30a in the left ventricular muscle of the 0h group after exercise were significantly increased compared with those of the quiet control group, Beclin-1 mRNA and LC3 mRNA (P\u003c0.05). The expression also increased significantly (P\u003c0.05), and did they all return to a quiet level after 3 hours of exercise?
Conclusion: Is it acute exercise myocardial ACE mRNA? miRNA30a? Beclin-1 mRNA and LC3 mRNA can be significantly increased. Said that this temporary increase may be caused by exercise stress?
