【Animal Experiment】-The effect of fasting time on glucose tolerance and insulin secretion in Kunming mice

Purpose: In certain environments, to ensure that Kunming mice, the most used in China, are relatively stable in OGTT?

Method: Oral glucose tolerance test in mice (Oral Glucose Tolerance Test, OGTT) is to study the function of β cells in the island and the ability to regulate blood sugar? Fasting is a necessary step before the experiment, but when the fasting time is different, will fasting and glucose stimulation cause the blood sugar level of mice to increase? Are there specific differences in insulin levels? In this experiment, 6-7 weeks were selected, and old clean grade male mice in Kunming were randomly divided into 4 groups, fasted for 4-8-8-12 hours, and then each group was subjected to OGTT. Tend to detect changes in blood sugar and insulin levels and analyze changes in blood sugar and insulin levels?

Result: (1) The blood glucose and insulin levels of all mice rose from the fasting baseline to the critical level, and then dropped to the baseline. Compared with the fasting blood glucose levels of mice fasted for 4 hours, the fasting blood glucose levels of the other three groups of mice in the fasting group were significantly lower than the fasting blood glucose levels of fasting for 4 hours (4 hours). P\u003c0.01)? (2) From the perspective of blood glucose, the 10-hour blood glucose concentration of the fasting group rose the most from the fasting threshold and its blood glucose curve. Compared with the 4-hour fasting group, is there a significant difference in the area below the fasting group (P\u003c0.05)? (3) From the viewpoint of insulin secretion, the fasting insulin concentration for 4 hours is 4% higher than the 8 hour fasting insulin concentration of about 12 hours for fasting insulin. After doubling and fasting for 8 hours, will the area under the insulin curve be minimized?

Conclusion: Are the blood glucose and insulin levels of Kunming mice fasted for 8-10 hours with OGTT significantly different? High resolution and better trends, more reasonable time allocation during fasting, is the experimental plan reasonable? high efficiency. Can the fasting time of the glucose tolerance test be used as a reference when using Kunmei mice to establish a diabetes model in the future?
