[Animal Experiment]-Effect of Shenren Huoxue Granules on the TGF-β1/Smad signaling pathway in rat liver fibrosis induced by carbon tetrachloride

  Objective: To observe the effect of Shenren Huoxue Granules on the TGF-β1/Smad signaling pathway of rat liver fibrosis induced by carbon tetrachloride, and to explore the mechanism of Shenshen Huoxue Granules in treating liver fibrosis?

  Method: 50 male SD rats were randomly divided into normal, model, low, medium and high doses of Shenren Fushe Granules. The normal group was intraperitoneally injected with normal saline 1mL/kg, and the model group and each group were given 40% carbon tetrachloride. Twice a week, twice a week, 0.2mL/100g intraperitoneal injection each time, for 8 consecutive weeks, the low-dose group takes 1.575g/(kg?bw) every day from the 4th week, and the medium-dose group is 3.15g. From the 4th week/day (kg? bw)? In the high-dose group, 6.3g/(kg·bw) was intragastrically administered for 4 weeks, and the rats were killed at the 8th week.   Result: Compared with the normal group, the liver lobule structure of the model group was damaged. Although the fibers increased significantly and formed pseudolobules of various sizes, the degree of liver fibrosis in each dose group of Shenren Huoxue Granules was significantly higher than that of the model group, and the height of Shenren Huoxue Granules was higher. Is the dose group the most effective? The expression of model rat liver collagen I, collagen III, TGF-β1 and Smad3 was significantly higher than that of the normal group (P\u003c0.05), and the collagen I, collagen III, and TGF in the human Huoxue Granules in each dose group -β1 and Smad3 are higher than the normal group. The expression of Smad7 was higher, but the expression of Smad7 was significantly reduced (P\u003c0.05). The model group decreased (P\u003c0.05), and the expression of Smad7 was higher than that of the model group. The high-dose group Shenren Huoxue Granules is the most effective (P\u003c0.05)?

  Conclusion: Shenren Huoxue Granules can significantly improve liver fibrosis in rats. Is the mechanism of TGF-β1 in rat liver tissues? Could it be related to the ability to down-regulate the expression of Smad3? Up-regulate the expression of Smad7, thereby up-regulating collagen I? Collagen III?
