【Animal Experiment】-Wubao Capsule's effect on Th1/Th2, Th17/Treg imbalance and chemokines in alveolar lavage fluid of asthmatic mice

Objective To observe the effect of national medicine Wubao Capsule on Th1/Th2, Th17/Treg8, Iota, and chemokines in the alveolar lavage fluid (bronchoalveolavagefluids, BALF) in asthmatic mice induced by ovalbumin (OVA) The influence of MCP~1.

Methods BALB/c mice were randomly divided into 5 groups (normal group, model group, dexamethasone group, Wubao capsule low and high dose groups). The asthma model was replicated by OVA sensitization and challenge methods. He and PAS staining was observed. Histopathological changes in mouse lungs, rapid Wright-Giemsa staining method to observe the number of various types of inflammatory cells in BALF, ELISA determination of IFN ̄γ, IL ̄4, IL ̄17A, IL ̄10, Eotaxin, IL ̄ in mouse BALF 8. The content of MCP~1.

Results Wubao Capsules can significantly reduce the inflammatory infiltration and mucus secretion in the lung tissues of asthmatic mice, reduce the number of various types of inflammatory cells and the production of IL-4, IL-17A, Eotaxin, IL-8, and MCP-1 in BALF (P< 0.01, P<0.05), and can significantly increase the content of IFN ̄γ and IL ̄10 (P<0.01, P<0.05).

Conclusion Wubao Capsules may play a role in preventing and treating asthma by regulating the balance of Th1/Th2, Th17/Treg-related cytokines in BALF and inhibiting the production of chemokines Eotaxin, IL-8, and MCP-1.
