Objective: To explore the protective effect and mechanism of activated protein C (APC) on myocardial ischemia-reperfusion injury in rats.
Method: 30 male SD rats were randomly divided into sham operation group (10), I/R group (10) and APC treatment group (10). The left anterior descending branch was ligated for 60 minutes and reperfused for 6 hours. The myocardial ischemia-reperfusion model was established. ELISA method and immunohistochemical method were used to detect the expression levels of TNF-α, myocardial tissue MPO and ICM-1, and HE staining method was used to detect the pathological changes of each group. detected. Results Compared with the sham operation group, the expression levels of TNF-α, MPO and ICM-1 in the I/R group were significantly increased (P\u003c0.01). Compared with TNF-α in the APC group, the expression levels of α, MPO and ICM-1 were significantly lower (P\u003c0.01), and the infiltration of inflammatory cells was observed under a light microscope. The I/R group is the most obvious. Compared with the IR group, the APC group has a significant decrease.
Conclusion: APC has a protective effect on myocardial ischemia-reperfusion injury, and its mechanism may be related to inhibiting the expression of inflammatory factors and reducing tissue inflammatory cell infiltration.
