【Animal Experiment】-Study on the anti-tumor effect of Xiuzhen mushroom powder on H22 tumor-bearing mice

Objective This test is to study the anti-tumor effect of Xiuzhen mushroom and its anti-tumor mechanism.

Methods 60 male ICR mice were selected, 10 were selected as the blank group, and the remaining 50 mice were subcutaneously inoculated with H22 mouse ascites to construct a tumor-bearing mouse model. After modeling, they were randomly divided into model group and positive drug (CTX ) Group, Xiuzhen mushroom low, medium and high dose groups, each group has 10. The mice in the model group were given normal saline, the mice in the positive drug group were injected intraperitoneally with cyclophosphamide (CTX) physiological saline solution at 20 mg/kg body weight every other day, and the mice in the low, medium, and high dose groups of Xiuzhen mushroom were injected with 750 mg/kg body weight. Daily doses of 1500 and 3000 mg/kg body weight were administered with Xiuzhen mushroom physiological saline suspension. After 10 days of administration, the average tumor weight and tumor inhibition rate of mice in each group were determined; the index of immune organs, serum immunoglobulin and cytokine content were determined; the antioxidant indexes of liver and kidney were determined; the HE staining of tumor and spleen tissue was observed Pathological section.

Result (1) The average tumor weight of CTX and Xiuzhen mushroom low, medium and high dose group mice were all extremely significantly lower than the model group (P<0.01), and the tumor inhibition rates of the four groups of mice were 55.18%, respectively. 29.06%, 47.47% and 48.80%.

  (2) Compared with the blank group, the spleen index, serum IgA and TNF-α content and liver MDA content of model group mice were significantly increased (P<0.05, P<0.01), and="" the="" serum="" il-6="" content="" increased="" trend="" p="">0.05), and serum IL-2 content, liver CAT and GSH-Px activity and kidney SOD, GSH-Px and CAT activity significantly reduce (P<0.05, P<0.01); Simultaneously The thymus index of mice in the CTX group was significantly lower than that in the blank group (P<0.05).

  (3) Xiuzhen mushroom has a reversal effect on the abnormal changes in immune and antioxidant functions of tumor-bearing mice: Compared with the model group, the serum IgA and TNF-α levels of mice in each dose group of Xiuzhen mushroom and IL-6 levels of mice in the high-dose group Significantly reduced (P<0.05, P<0.01), the serum IL-2 level of mice in the low-dose group was significantly increased (P<0.01); Xiuzhen mushroom treatment also significantly increased the tumor-bearing cells in each dose group Rat liver CAT activity and kidney SOD and CAT activity (P <0.05, P <0.01), significantly increased the liver GSH-Px activity in the middle and high dose groups and the kidney GSH-Px activity in the low and middle dose groups (P <0.01) 0.05, P <0.01), and significantly reduced the liver MDA content in the middle dose group (P <0.05).

  (4) Compared with the CTX group, the spleen index of the mice in the Xiuzhen mushroom high-dose group was significantly increased (P<0.05), while the serum IL-6 level was significantly reduced (P <0.05).

  (5) The tumor histopathological section showed that the area of tumor necrosis in the CTX group and the Xiuzhen mushroom each dose group increased significantly.

Conclusion Xiuzhen mushroom can inhibit the growth of H22 solid xenograft tumors, and its mechanism is related to its strong immune regulation and antioxidant effects.
