【Zhonghong Boyuan】-Establishment of acute and chronic hyperuricemia animal models

      Objective: To establish acute and chronic hyperuricemia models in rats, use positive drugs to verify the sclerosis of the two models, and provide model tools for the development of anti-hyperuric acid drugs.

  Method: A combination of intraperitoneal injection of hypoxanthine and subcutaneous injection of potassium oxazine was used to simulate acute gout model mice. Allopurinol tablets were administered to the positive group 30 minutes after model building, and uric acid and blood creatinine, serum urea nitrogen and hepatic xanthine oxidase (XOD) activities were measured 2 hours after model building. Rats with chronic gout were injected with adenine and ethambutol hydrochloride every day to establish a model. The positive group received allopurinol tablets every day. After 21 consecutive days, blood uric acid, blood creatinine, serum urea nitrogen, liver XOD activity and renal tissue diseases were detected.

Results: The blood uric acid and blood creatinine of acute and chronic gout model animals were significantly increased (P\u003c0.05), and the serum urea nitrogen and liver XOD activity of chronic gout rats were also significantly increased ((P\u003c0.05) 。 Quality damage and deterioration of uric acid crystals, the score increased significantly (P\u003c0.05). The positive drug allopurinol can significantly reduce the blood uric acid level of acute and chronic hyperuric acid animals, and reduce the blood creatinine level and blood creatinine levels of chronic hyperuric acid animals. Liver XOD activity, improve kidney damage, and improve uric acid levels. Reduce salt crystals (P\u003c0.05).

   Conclusion: The method used in this experiment is suitable for the establishment of acute and chronic hyperuricemia animal models, and can be used to observe the pharmacodynamics of drugs.
