【Animal Modeling】-Cobalt-induced chronic epilepsy model

      1. Model material: rat; drug: pentobarbital, cobalt powder, kanamycin; instrument: stereotaxic device.

      2. Modeling method Anesthetize model rats by intraperitoneal injection of pentobarbital 30 mg/kg, fix the pentobarbital 3 mm behind the front line, and fix it on the right side of the midline, and use rongeurs to cut the 8 mm diameter skull. Then, cut open the dura mater. Place about 30 mg of sterile cobalt powder, about 10 mg2, on the front surface of the cortical motor area, connect the recording electrode and suture. Kanamycin 125 mg / (only 1 day), intramuscular injection for 3 consecutive days to prevent infection.

   3. Modeling principle Cobalt can cause chronic epilepsy in animals.

  4. The change after modeling is usually 2-3 weeks after placing the cobalt powder. There are fusiform spores on the contralateral limb of the placed cobalt. In some animals, epileptic seizures generally occur after localized fusiform spores, and the intensity of the seizures gradually increases from 4-6 weeks. Weaken or disappear. Placing cobalt in the limbic system of cobalt, such as the amygdala or hippocampus, can cause neuromotor epileptic seizures. Animals exhibit reduced activity, fear, shortness of breath, and constant grasping.

   5. Precautions Sterilize surgical instruments to prevent surgical infections, surgical trauma should be as small as possible, and strictly aseptic operation. After the operation, the rat can be separated and awakened for 3-5 hours, and then placed in a cage to prevent licking or biting the wound of the unconscious rat during the first awakening. In the breeding room, try to keep the temperature and humidity of the rats at their best.
