【Animal Experiment】-The effect of the venous blood cell suspension storage time on the blood routine measurement of mice

  Objective: To observe the effect of the preservation time of venous blood cell suspension on routine blood routine of mice.

  Method: 20 adult ICR mice and C57BL/6J mice were randomly selected, and quantitative venous blood was collected, added to the diluent for quantitative blood cell analysis, and mixed thoroughly for 0 minutes and 15 minutes. Prepare the venous blood cell suspension in minutes, 30 minutes and 1 hour. , 2 hours, 4 hours, .6 hours, 8 hours, the MEK-6410C automatic hematology analyzer uses micro hematometry to determine blood routines. Results The blood routine data changes of ICR mice and C57BL/6J mice were basically the same. Compared with 0 minutes, RBC decreased significantly at 2, 4, 6, and 8 hours (P0.05).

  Conclusion: When microblood measurement has confirmed the blood routine of mice, it should be completed as soon as possible after completing the venous blood cell suspension. The total number of red blood cells should be determined within 1 hour. The measurement of HCT and MCV indicators related to RBC should not exceed 30 minutes. The PLT decision should be completed within 2 hours. PCT and MPV decisions related to PLT should be made within one hour. The total number of WBC and HGB content is relatively stable. It was completed in 8 hours.
