[Animal experiment]-Qingfei Touxie Decoction mediates NLRP3 inflammasome to inhibit NF-κB signaling pathway and improves inflammatory response in mice with Mycoplasma pneumoniae

  Objective: To explore the therapeutic effect and mechanism of Qingfei Touxie Decoction on mycoplasma pneumonia (MPP) mice.

  Method: 40 male SPF BALB/c mice were randomly divided into control group, model group, Qingfei Touxue Decoction low, medium, and high dose groups, each with 8 mice. A mouse model of Mycoplasma pneumoniae pneumonia was established by intranasal injection of Mycoplasma pneumoniae (MP). The mice in the model group were given physiological saline. The Qingfei Touxie Decoction suspension was used to administer 800 mg/kg body weight daily. After 2 weeks of treatment, the mouse lung tissue index was calculated. HE staining is used to detect pathological changes in lung tissue. TUNEL is used to detect apoptosis in mouse lungs. ELISA was used to detect IL-1β, TNF-α and IL in the alveolar lavage fluid of each mouse. -6, IL-18 content; qPCR to detect the expression of NLRP3, MyD88 and NF-κB mRNA in lung tissue; Western blotting to detect the expression of NLRP3, MyD88 and p-NF-κB protein in lung tissue.

  Results: Qingfei Touxie Decoction can significantly reduce the lung index of MPP mice, reduce lung pathological damage and cell apoptosis rate (P\u003c0.05); Qingfei Touxie Decoction can be used for IL-1β in alveolar lavage fluid And TNF was also significantly reduced. -α, IL-6, IL-18 levels (P\u003c0.05) and NLRP3, MyD88, NF-κB (P\u003c0.05) mRNA and protein expression decreased.

  Conclusion: Qingfei Touxue Decoction can improve lung injury in mice with Mycoplasma pneumoniae pneumonia, and its mechanism of action may be related to the inhibition of NLRP3 matrix and NF-κB signaling pathway.
