Objective: To investigate the inhibitory effect of Zhenqi Fuzheng Granules on angiogenesis in liver cancer model rats and its possible mechanism.
Methods: Wistar rats were divided into control group (control group), model group (prepared by low-dose diethylnitrosamine (DEN) induction method), Zhenqi Fuzheng Granules group (2.62g/kg) and si-miR- . In the 200c group (20pm), 12 rats in the Zhenqi Fuzheng Granule + si-miR-200c group were treated for 8 weeks in each group. HE staining to observe the pathological changes of rat liver tissue; enzyme-linked immunoassay to detect serum vascular endothelial growth factor (VEGF), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels; real-time fluorescence Quantitative PCR method to detect the expression of miR in liver tissue 200c; Western blot method to detect the expression of zinc finger protein (ZEb-2) and VEGF protein in liver tissue; ZEb-2, VEGF and immunohistochemical method to detect microvessel density (MVD) positive expression.
Result: Compared with the model control group, the VEGF, ALT and AST levels of the model group increased, and the number of aeocytes and necrotic cells in the liver tissue increased. The expression of miR-200c in liver tissue decreased, and the expression of ZEb-2 and VEGF protein increased. The positive rate of ZEb-2, VEGF and MVD increased (P\u003c0.05). Compared with the model group, the levels of VEGF, ALT and AST in the Zhenqi Fuzheng Granule group decreased, the degree of cell deformation decreased, the degree of differentiation of cancer cells increased, and the expression of miR-200c increased. , ZEb-2 and VEGF protein expression decreased, and ZEb-2 expression decreased. In the si-miR-200c group, the positive expression rate of VEGF and MVD decreased, the levels of VEGF, ALT and AST increased, the degree of cell carcinoma increased, the expression of miR-200c in liver tissue decreased, the expression of ZEb-2 and VEGF protein increase. 2. The positive rate of VEGF and MVD increased (P\u003c0.05). Conclusion: Zhenqi Fuzheng Granules can inhibit angiogenesis in liver cancer model rats and reduce canceration. This mechanism may be related to the up-regulation of miR-200c expression and subsequent negative regulation of the expression of ZEB-2 protein.
