Objective: To explore the effect of a high-fat diet of sesame oil rich in n-3 polyunsaturated fatty acid (PUFA) on insulin sensitivity and related gene expression in insulin-resistant rats.
Methods: Insulin resistance model rats were randomly divided into 2 groups: high fat group (HF) and perilla oil group (PO). PO was 20% of the ratio of lard oil in HF instead of perilla oil. %, 4 weeks later, the insulin sensitivity of rats was measured; gas chromatography was used to detect the content of α-linolenic acid (α-linolenic acid, ALA) in rat serum; Western blot was used to detect skeletal muscle glucose transporter 4 4, GLUT4) and insulin receptor substrate-1 (insulin receptor substrate-1, IRS-1) protein expression; real time PCR method to detect skeletal muscle Glut4, Irs-1 mRNA expression.
Results: After 4 weeks of intervention with sesame oil, compared with the HF group, with the increase in ALA intake and serum ALA content in the PO group, the expression of GLUT4 protein and mRNA increased significantly, while the IRS-1 protein and The mRNA expression was significantly reduced; the results of the hyperinsulinemic-hyperglycemic clamp experiment showed that there was no significant difference in insulin sensitivity between the two groups of rats.
Conclusion: ALA (0.556 g/d) high-fat diet intervention can regulate the expression of skeletal muscle GLUT4 and IRS-1, but it cannot improve insulin sensitivity in insulin-resistant rats.
