Animal experiment: the effect of ovariectomy on skin aging in osteoporotic rats

  Purpose: The osteoporosis model constructed with ovariectomized rats is the most suitable model for studying postmenopausal osteoporosis. However, there is very little research on the effect of rat ovaries on the skin. The purpose of this study is to observe the effect of ovariectomy on the skin aging indicators of rats.

  Methods: Twenty three-month-old healthy SD female rats were randomly divided into sham operation group (Sham group) and ovariectomized group (OVX group). Three months after the removal of the ovaries, serum estradiol and skin tissue levels were measured. Aging-related biochemical indicators such as catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activity and malondialdehyde (MDA), hydroxyproline ( HYP), lipofuscin (LF) content, and observe the pathological changes of skin tissues, and use computer image analysis system for quantitative analysis.

  Results: The level of serum estradiol in the OVX group was significantly reduced; histopathological observation showed that the thickness of the epidermis and dermis of the OVX group was significantly thinner than that of the Sham group (P<0.01), the skin collagen fibers were reduced, and the arrangement was loose; compared with the Sham group , The activities of CAT, GSH-Px and SOD in the skin of rats in the OVX group were significantly reduced (P<0.01), and the content of MDA was significantly higher than that in the Sham group (P<0.01), the content of HYP in the skin tissue was significantly reduced, and the content of LF was significantly increased (P< 0.01).

  Conclusion: 3-month-old rats can cause skin aging after 3 months after ovarian removal, which may be a suitable model for studying postmenopausal skin aging.
