Objective: To study the effect of different pH drinking water on the gut microbial diversity of SPF mice.
Methods: Ninety 21-day-old SPF mice were randomly divided into 3 groups, 30 in each group, and were given three types of water, namely acidified water (pH 3.0), neutral water (pH 7.0) and alkaline water (pH 9.0). And for three months of breeding. In each group, 10 animals died in 4 weeks, 8 weeks, and 12 weekends. Serum was collected to detect bacteria, viruses, and parasites according to SPF national standard items. The blind contents were retrieved, and primers were designed according to the conservativeness of bacterial 16S rRNA. , Construct a library, perform high-throughput (Illumina) sequencing, obtain 10 G data, and perform statistics and cluster analysis on the data.
Result: The intestinal microbes of mice treated with pH 3.0 have moderate abundance and high stability.
Conclusion: Under the conditions of large-scale breeding, selecting pH 3.0 drinking water mice can maintain the diversity of intestinal microbes for a long time, and the stability reaches a plateau. This study provides theoretical and technical support for the long-term stable production practice of maintaining animal quality, and is also a specific application of gastrointestinal metagenomics technology in the field of animal quality control.
