Objective: To observe the effect of Shenren Huoxue Granules on the TGF-β1/Smad signaling pathway of liver fibrosis in rats induced by carbon tetrachloride, and to explore the mechanism of Shenren Huoxue Granules in treating liver fibrosis.
Methods: Fifty male SD rats were randomly divided into normal group, model group and Shenren Huoxue Granules low, medium and high dose group. The normal group was given intraperitoneal injection of normal saline 1mL/kg, and the model group and Shenren Huoxue Granules were given each group. 40% carbon tetrachloride 0.2mL/100g intraperitoneal injection, 2 times a week, for 8 consecutive weeks, from the 4th week onwards, Shenren Huoxue granules were given respectively, and the low-dose group was 1.575g/(kg∙bw) daily, medium dose Group 3.15g/(kg∙bw) daily, high-dose group 6.3g/(kg∙bw) daily, gavage for 4 consecutive weeks, sacrifice the rat at the 8th week. HE staining and Masson staining to detect rat liver Fibrosis degree; immunohistochemical detection of rat liver collagen I, collagen III, TGF-β1 expression; real-timePCR and Western blot detection of rat liver TGF-β1, Smad3, Smad7 expression.
Results: Compared with the normal group, the structure of liver lobules was damaged in the model group, and the fibers were significantly increased, forming pseudo-lobules of different sizes; while the level of liver fibrosis in each dose group of Shenren Huoxue Granules was significantly improved compared with that of the model group. The high-dose group has the most significant curative effect. ②The expression of collagen I, collagen III, TGF-β1, and Smad3 in the liver of the model group was significantly higher than that in the normal group (P<0.05), and the expression of Smad7 was significantly reduced (P<0.05). The expression of collagen I, collagen III, TGF-β1, and Smad3 in each dose group of Huoxue Granules was lower than that of the model group (P<0.05), and the expression of Smad7 was higher than that of the model group. The high-dose group of Shenren Huoxue Granules had the best effect (P<0.05). ).
Conclusion: Shenren Huoxue Granules can significantly improve liver fibrosis in rats, and its mechanism may be related to its ability to down-regulate the expression of TGF-β1 and Smad3 in rat liver tissues and up-regulate the expression of Smad7, thereby reducing the synthesis of Collagen I and Collagen III.
