Comparison of depression and anxiety-like behaviors in mice of different ages

Objective To observe and compare the depression and anxiety-like behaviors of experimental mice at different ages.

Methods 27 male C57BL/6 mice were divided into young group (3 months old, n = 9), middle-aged group (10 months old, n = 9), and elderly group (18 months old, n = 9) according to their age. The tail suspension test, the forced swimming test, the elevated plus maze test, the open open field test and the sugar water preference test were performed respectively to observe depression and anxiety-like behaviors in mice, and the liquid mass spectrometry (LC-MS/MS) method was used to detect C57BL/ 6 The serum serotonin content of mice.

Results The tail suspension time of mice in the middle-aged group was longer than that of the old group mice (P<0.05); the forced swimming immobility time of the young group, middle-aged group, and old group mice decreased significantly, and the young group mice swimming The immobility time was significantly longer than that of the middle-aged group of mice (P<0.001) and the old group of mice (P<0.001), and the swimming immobility time of the middle-aged group of mice was significantly longer than that of the old group of mice (P<0.01). 01); the middle-aged group of mice has a higher 12-hour sugar preference rate than the old group (P<0.05), and the young group has a higher 12-hour, 36-, 48-, and 60-hour sugar preference rate than the old group (P<0.05). <0.05); the old group of mice has a longer open arm time than the middle-aged group and the young group of mice (P<0.05), and the young group of mice has more probe times than the middle-aged group of mice (P<0.01). ); The total distance of movement of the young group of mice in the open field is longer than that of the old group of mice (P<0.05); the serum 5-HT content of the old group, the middle-aged group, and the young group showed a decreasing trend, and the young group The serum 5-HT of mice was significantly lower than that of the middle-aged group (P<0.05) and the elderly group (P<0.01).

Conclusion On the whole, under the same stimulus, the younger the mouse is, the more likely it is to show depression and anxiety-like behavior, and the lower the serum 5-HT level.
