Objective To observe the effect of venous blood cell suspension storage time on blood routine measurement in mice.
Methods Twenty adult ICR mice and 20 C57BL/6J mice were randomly selected. Quantitative venous blood was collected and added to the diluent for quantitative blood cell analysis to make a venous blood cell suspension at 0min, 15min, 30min, 1h, 2h , 4h, .6h, 8h on the MEK-6410C automatic blood cell analyzer using micro blood assay to determine blood routine.
"Results ICR mice and C57BL/6J mice blood routine data change trend is basically the same. Compared with 0 min, RBC was significantly reduced at 2h, 4h, 6h, and 8h (P<0.01), PLT was significantly increased at 4h, 6h, and 8h (P<0.01), and PCT was significantly reduced at 2h, 4h, 6h, and 8h. Significantly increased (P<0.01), MPV increased significantly at 2h, 4h, 6h and 8h (P<0.01), MCV decreased significantly at 1h, 2h, 4h, 6h and 8h (P<0.01), MCHC Significantly increased at 30min, 1h, 2h, 4h, 6h and 8h (P<0.01), MCH increased significantly at 1h, 2h, 4h, 6h and 8h (P<0.01)), HCT at 1h, 2h , 4h, 6h and 8h were all significantly reduced (P<0.01), pdw="" and="" hgb="" had="" no="" significant="" changes="" p="">0.05).
Conclusion When the microblood measurement method measures the blood routine of mice, it should be completed as soon as possible after the venous blood cell suspension is made. The total number of RBCs should be determined within 1 hour. The determination of HCT and MCV indicators related to RBC should not exceed 30 minutes. The determination of PLT should be completed within 2 hours. The determination of PCT and MPV related to PLT should be within 1 hour. The total number of WBCs and HGB content are relatively stable. Complete within 8h.
