Purpose: To construct a rat model of type 2 diabetes with coronary heart disease.
Method: A Zucker diabetic fatty (ZDF) model of type 2 diabetes with coronary heart disease was prepared by feeding with high-fat diet combined with continuous injection of isoproterenol hydrochloride (ISO). The model was evaluated by serum creatine kinase (CK), serum creatine kinase isoenzyme (CK-MB), electrocardiogram ST and heart tissue morphology.
Results: The expression of CK in the non-DM model group and the DM-CHD group gradually increased with the increase in the number of administrations, and the expression of CK-MB first increased and then decreased, indicating that myocardial injury in rats has formed. The deviation of ST segment in lead II of the model rat ECG indicates that myocardial injury has been formed. The myocardial pathological section results of the rats in the injection model group showed that the myocardial cells of the blank control group were normal. The myocardial necrosis area of the ZDF rats reached 1/2 of the myocardial cross section after being continuously injected with ISO for 5 days, and the myocardial necrosis area reached the myocardial cross section after 10 days. 3/4 of it.
Conclusion: CK, CK-MB content changes, ECG ST wave changes and myocardial pathological slices show that the model has been successful. ISO can cause a certain degree of myocardial damage. Continuous injection of ISO can be used to construct a coronary heart disease model. In this paper, a simple method for constructing a rat model of diabetes and coronary heart disease with high-fat feed and continuous injection of ISO was established.
