Study on the optimization plan of the animal model of Ureaplasma urealyticum infection

  Purpose: To investigate the advantages and disadvantages of various modeling methods, and to select the most suitable animal model method for Staphylococcus aureus infection. Methods: SPF-grade BALB/c female mice and Wistar rats were selected to establish a low-dose UU3 bacterial solution for multiple infections, a high-dose UU3 bacterial solution for one infection, and a large dose of estradiol benzoate for the solution Alcohol pretreatment was used as an intervention. Animal models infected with Ureaplasma on the 14, 28, 42, and 56 days after the initial vaccination, the cervical secretions of experimental animals were collected in Ureaplasma cultures, and the reproductive organs were collected for morphological analysis.

  Result: ①Compared with Wistar rats, BALB/c mice have poorer general conditions, with increased vaginal secretions and genital ulcers, but there is no significant change in the genital area of Wistar rats. (2) The comparison of the formation rate of UU colony in the vagina of rats in each group was statistically significant (P\u003c0.05). Compared with the low-dose estrogen group and the low-dose non-estrogen group, the difference was statistically significant (P\u003c0.05). The difference between the estrogen group and the high-dose non-estrogen group was statistically significant (P\u003c0.05). The difference between the low-dose estrogen group and the high-dose estrogen group was statistically significant (P\u003c0.05). There was a statistically significant difference between the estrogen group and the high-dose non-estrogen group (P\u003c0.05). The results of each group of mice are similar to those of rats. ③Under the same intervention, there were statistical differences between rats and mice treated with low-dose estrogen and between rats and mice treated with high-dose estrogen (P\u003c0.05). There is a statistically significant difference (P\u003c0.05). P\u003c0.05), indicating that the vaginal UU colony formation rate of mice is higher under the same treatment. ④ Visual observation: The reproductive organs of the rats in the low-dose estrogen group had edema, the other group of rats had good flexibility to the gonads, and the control group had good flexibility to the genital tissues. The low-dose estrogen group had sex. The genital tissues of the mice were hyperemia, hydrosalpinx, thickened, stiffness and decreased elasticity; the mice in the high-dose estrogen group were mainly edema. The low-dose non-estrogen group and the high-dose non-estrogen group were only seen in the vagina and neck, with mild congestion and edema. (5) Microscopic image: There was no obvious pathological damage to the vaginal tissue of rats in each intervention group, and tissue relaxation and edema were observed. The mice in the low-dose estrogen group had severe pathological damage; the low-dose non-estrogen and high-dose estrogen mice had less congestion and edema, slightly larger glands, and inflammatory secretions.    Conclusion: Compared with Wistar rats, BALB/c mice have obvious genital damage. Estrogen pretreatment may increase the rate of UU colonization. BALB/c mice have a high rate of UU colony formation in the vagina, while BALB/c mice have severe pathological damage, which is more valuable for animal experiments.
