Objective: To investigate the expression pattern of TET family proteins in mouse lung cancer models.
Method: Divide mice into 3 groups, each with 10 mice. Set as control group (Con), urethane induction group (Ure), and hydroxyurea administration group (Ure-HU). Ure group and Ure-HU group were given 800mg/kg uratan solution for 10 weeks to induce the establishment of mouse lung cancer pattern. After 10 weeks, HU drug (500mg/kg) was given to Ure-HU group for 21 days. Observe the number of lung nodules. The expression patterns of TETs in the 3 groups of mice were analyzed by qPCR and Western blot.
Result: Compared with the Con group, the Ure and Ure-HU groups lost significant weight. Morphological observation showed that both Ure group and Ure-HU group had lung tissue lesions. The qPCR results showed that in the Ure group, the expression of TETs was significantly reduced (P<0.05). After HU treatment, the expression of TET1 and TET2 in Ure-HU group increased (P <0.05). Westernblot and qPCR results are consistent.
Conclusion: In the mouse lung cancer model induced by urethane, the expression pattern of TETs protein is abnormal, which proves that TET1 and TET2 play an important role in the process of lung cancer.
