Comparative study on establishing mouse models of allergic conjunctivitis under different conditions

  Objective To establish a mouse model of allergic conjunctivitis under the conditions of different antigens, different excitation pathways, and different ages of mice, and to compare the immune response status to screen the best sensitization conditions. Establish and provide new ideas and methods. Methods 65 SPF Balb/c mice were exposed to different antigens (ovalbumin, ragweed pollen, house dust mites), different excitation routes (gavage, nebulization, subcutaneous injection), and different mouse ages (1 A mouse model of allergic conjunctivitis was constructed under the conditions of one week, two weeks, three weeks, and eight weeks. RT-qPCR was used to detect the relative expression levels of CCL5 mRNA and IL-17 mRNA in mouse conjunctival tissues, and flow cytometry was used to detect the percentage of IL-17 in splenic single cell suspension in CD4+ T cells.

   Results Compared with the negative control group, the percentage of IL-17, the expression of CCL5 mRNA and IL-17 mRNA in the ovalbumin group, house dust mite group, and ragweed pollen group all increased. Among them, the ragweed pollen group had the most significant increase. Compared with the negative control group, the percentage of IL-17 and IL-17 mRNA expression in the subcutaneous injection group, gavage group, nebulized inhalation group, and positive control group increased. The expression of CCL5 mRNA in the gavage group was similar to that of the negative control group. The expression level of the subcutaneous injection group was closer to the positive control group. The percentage of IL-17, CCL5 mRNA and IL-17 mRNA expression levels of mice in the 1-week and 2-week-old groups were similar, but significantly lower than those in the 3-week-old and 8-week-old mice.

   Conclusion Ragweed pollen has better allergenicity in constructing mouse animal models of allergic conjunctivitis. Subcutaneous injection to stimulate antigen can achieve satisfactory results in the structure of allergic conjunctivitis mouse animal model. Although there are no obvious ocular symptoms and signs, the whole body and local are all sensitized. The immune response of 3-week-old Balb / c mice is closer to that of adult Balb / c mice.
