Objective To study the therapeutic effect of Huangqi Sanqi Mixture on diabetic nephropathy (DN) mice and explore the potential regulatory mechanism. Methods Fifty C57BL/6 male mice were randomly divided into control group (NC group), model group (DN group), Huangqi Sanqi mixture low/high dose group (L-A&P, H-A&P group) and Irbesartan Group (IRB group). The NC group was given ordinary feed, and the rest of the groups were fed with high-fat and high-sugar feed for 2 months. After 5 days of continuous intraperitoneal injection of streptozotocin 50 mg/(kg·d) to induce diabetic nephropathy model, different gavage was given after modeling Treatment, the mice were sacrificed after 4 weeks of treatment. Renal pathology was observed by HE and PAS, and renal function and inflammatory factors (TNF-α, IL-1β, IL-6), lncRNA Arid2-IR, NF-κB in mouse kidney tissue were detected And the expression of downstream signaling molecules. Results Compared with the DN group, the 24-hour urine protein, serum creatinine and urea nitrogen of the mice after A&P treatment decreased significantly, and the renal function was restored to a certain extent; the renal pathological structure was significantly improved, and the inflammatory factors (TNF-α, IL- The expression of 1β, IL-6) decreased in a dose-dependent manner, and the expression of Arid2-IR, NF-κB and its downstream molecules (COX2, IL-6) were down-regulated, indicating that the activation of NF-κB signaling pathway was inhibited.
Conclusion A&P can effectively alleviate the renal inflammation in DN mice, and its mechanism may be related to the regulation of the Arid2-IR/NF-κB signal axis.
