The key to mouse osteosarcoma model construction

Objective: To explore the key factors for the success rate of tumor-bearing animal models of osteosarcoma.

Method: 24 C3h mice aged 4 to 8 weeks, including 6 C3h/HeJ type and 18 C3h/HeN type, were randomly divided into 2 groups. Group A used general depilatory cream to inject 0.2 mL of LM8 cell suspension in the lateral armpit after depilation; the concentration of LM8 cell suspension in the lateral armpit was 5×107 cells/mL after the animal shaver was used in group B; After 0.2 mL of LM8 cell suspension of order of magnitude/mL was given daily local massage at the injection site, and the tumor growth was recorded respectively.

Result: The rate of A-composition tumor is 33% (4/12); the rate of B-composition tumor is 100%.

Conclusion: Different injection techniques are of great significance to the tumor-bearing process of LM8 and the subsequent care for the tumor formation rate.
