Objective: To establish a rat model of rheumatoid arthritis combined with hypertension disease (arthritis-hypertension disease, AHD) and evaluate the characteristics of the model.
Method: Perform a surgical operation with a 0.25 mm silver clip to narrow the left renal artery of the rat to establish a hypertension (hypertension, HT) model, and at the same time inject 0.1 mL complete Freund's adjuvant into the left hind foot of the rat to establish an AHD model. The non-invasive blood pressure measurement and analysis system measures the tail artery pressure. The paw volume meter measures the swelling of the non-inflammatory side of the rat, and analyzes the arthritis index and the incidence of inflammation. The rats were sacrificed on the 35th day, and the thoracic aorta, ankle joint and spleen tissues were collected for HE staining and pathological examination.
Result: The joints of AHD model rats are obviously swollen, a large number of synovial cell proliferation, inflammatory cell infiltration, pannus formation in the joint cavity, an increase in the number of germinal centers in the spleen, a large number of lymphocyte infiltration, diffuse white pulp proliferation, and red pulp infiltration. Arthritis index, inflammation incidence, and histopathological scores of joints and spleen were significantly higher than those of adjuvant arthritis (AA) rats; at the same time, the blood pressure of AHD model rats was significantly increased, the thickness of the thoracic aorta and the media The cross-sectional area was significantly increased, the diameter of the lumen was significantly reduced, and the blood pressure and the degree of vascular damage were significantly higher or worse than those of HT rats.
Conclusion: Intradermal injection of complete Freund's adjuvant combined with surgical narrowing of the left renal artery can successfully establish a rat model of AHD.
