Improvement and evaluation of myocardial ischemia-reperfusion injury model in rats

  Purpose: To improve the preparation method of rat acute myocardial ischemia-reperfusion injury model, and conduct comprehensive evaluation, to lay a good model foundation for subsequent MI/RI research.

  Methods: Thirty-six clean Wistar rats were randomly divided into 3 groups: the normal group (Normal group), without intervention of the left anterior descending coronary artery (LAD); the sham operation group (Sham group), with only threading and no ligation; In the ischemia-reperfusion group (MI/RI group), the LD was ligated for 30 minutes and reperfused for 120 minutes. After the rats were anesthetized, the electrocardiogram was recorded continuously, the tracheotomy was performed with ventilator-assisted ventilation, and the rat’s LAD was intervened by the thoracotomy.   Results: The electrocardiogram of MI/RI group showed obvious ST-T dynamic changes; the level of creatine kinase isoenzyme (CK-MB) in MI/RI group increased significantly after 2 hours; compared with Sham group, the infarct area of MI/RI group Significant increase; the degeneration and necrosis of cardiomyocytes in MI/RI group were obvious, and inflammatory cells infiltrated extensively.

  Conclusion: The improved modeling method not only reduced the difficulty of surgery, but also successfully established a rat MI/RI model, laying a good model foundation for subsequent MI/RI research.
