Objective To try to use ovalbumin combined with smoking to establish a more
similar CVA model of guinea pigs.
Method A total of 24 SPF male and female guinea pigs were selected, and
they were randomly divided into a blank control group, an OVA sensitization
group, and an OVA sensitization + smoking group, a total of 3 groups, each with
8 guinea pigs. The OVA sensitization group was modeled by the ovalbumin
sensitization method, and the OVA sensitization + smoking group was modeled by
the ovalbumin sensitization combined with the smoking method. The rest methods
were the same. The lung compliance (Cydn), airway resistance (RI) after
methacholine challenge, the number of coughs and the time of first cough after
capsaicin challenge, lung tissue pathology and Ashcroft grading index changes in
the 3 groups were tested for evaluation. model.
Results Compared with the blank control group, the Cydn of the OVA
sensitization group and the OVA sensitization + smoking group significantly
decreased, the RI increased significantly, the number of coughs at 10 minutes
increased significantly, the first cough time was significantly earlier, the
lung tissue pathological changes were obvious, and the Ashcroft grading table
The score has risen significantly, and the above performances are consistent
with the characteristics of cough variant asthma. At the same time, compared to
the OVA sensitization group, the OVA sensitization + smoking group has better
performance in cough symptoms and changes in RI, and is more in line with the
characteristics of cough variant asthma. This may be related to reducing the OVA
sensitization concentration to avoid overreaction, Smoke increases cough
sensitivity is closely related.
Conclusion Using OVA sensitization combined with smoking can establish a
cough variant asthma guinea pig model with high similarity.
