Objective To observe and compare the effects of different genders and injection doses on the establishment of a rat model of myocardial hypertrophy induced by isoproterenol (ISO).
Method Take female and male SD rats respectively, and randomly divide them into three groups, normal group, low-dose group (1.25 mg/kg) and high-dose group (5 mg/kg). After 10 days of subcutaneous injection of ISO, the results of echocardiography, hemodynamics, histopathology, and real-time fluorescence quantitative PCR were compared under the influence of different factors.
Results (1) After high and low dose ISO treatment, the various parameters of rat heart weight, ventricular septal thickness, myocardial cell area, myocardial fibrosis degree, and hypertrophic genes (ANP, BNP) mRNA expression levels increased significantly (P<0 01), and the left ventricular function was significantly reduced (P <0.01); (2) male rats left ventricular weight/body weight, ventricular septal thickness, left ventricular end systolic pressure (LVESP), left ventricular maximum diastolic rate (-dp / dtmax) and the changes in ANP gene expression levels are in a significant dose-dependent relationship (P<0.01). Female rats only have left ventricular end systolic pressure (LVEDP) and left ventricular maximum diastolic rate (dp / dtmax) changes It is related to the dose, that is, the female low-dose group has a more significant reduction in cardiac function (P<0.01); (3) when low-dose ISO is injected, the reduction in cardiac function in female rats is more significant than that in male rats (P<0.01). 01); and when high-dose ISO was injected, the left ventricular weight/body weight and ANP gene expression increased more significantly in male rats than in female rats (P<0.01); (4) male rats survived in low and high dose groups The rates were 63.6% and 81.8%, respectively. Female low-dose and high-dose survival rates were 63.6% and 55.0%, respectively. There was no death in the normal group.
Conclusion Different ISO injection doses have a greater impact on the degree of myocardial hypertrophy in male rats, but have less impact on female rats. When modeling, male rats choose 5mg/kg dose, female rats choose 1.25mg/kg dose, the establishment of ISO-induced rat myocardial hypertrophy model effect is relatively good.
