Chronic myocardial ischemia mini-pig model with syndrome of phlegm and blood stasis-【animal modeling】

  Objective: To establish a miniature pig model of chronic myocardial ischemia with high-fat intravenous injection of vitamin D3 (VD3) and isoproterenol.

  Methods: Fifteen male Parmani piglets were randomly divided into a normal control group (Ctr), a high fat (HFC) group, a myocardial ischemia model (CMI) group, and a sputum-blood hyperemia (CMI) group. There are five animals in this group. The Ctr group is fed a normal diet, the HFC group is fed a high-fat diet, the CMI group is a high-fat diet, intravenous VD3 and isoproterenol, chronic venous and blood congestion syndromes are combined with the establishment of myocardial ischemia models. Measure animal body weight and electrocardiogram, respectively carry out 24 weeks of continuous modeling, activity, blood lipids, myocardial enzymes, hematology, inflammation, cardiac index (CI) and myocardial ischemic area. (MIS) Build models and evaluate the pathological process of hemorrhoids and hemostasis.

  Results: Compared with the Ctr group, the weight, heart rate (HR), total cholesterol (TC), low-density lipoprotein (LDL-C), high-density lipoprotein (HDL-C), atherosclerosis of the HFC group Index (AI), low, medium and high-cut whole blood and low viscosity, erythrocyte electrophoresis time (EPT), sensitive C-reactive protein (hs-CRP) and interleukin 6 (IL-) 6) levels were significantly increased (P\ u003c 0.05, P\u003c0.01); CMI group body weight, HR, 24 hour total ST segment increase (∑ST), average ST segment increase within 24 hours (ST_mean), activity, TC, triglycerides (TG), LDL-C, HDL-C, AI, creatine kinase (CK), lactate dehydrogenase (LDH), troponin 1 (cTn-1), low, medium and high cut whole blood and low viscosity , EPT, Carson viscosity (CV), hs-CRP, IL-6, CI and MIS all increased significantly (P\u003c0.05, P\u003c0.01), and adiponectin (APN) levels were significantly reduced ( P\u003c0.05). Compared with the HFC group, the AI, CK, LDH, cTn-1, low, medium and high cut whole blood viscosity, EPT, CI and MIS of the CMI group were significantly higher than those of the HFC group (P\u003c0.05, P\u003c0 .01), significantly lower than the APN level of the HPN group (P\u003c0.05). Correlation analysis shows that MIS is related to TC, LDL-C, AI, CK, LDH, cTn-1, APN, high, medium and low cut whole blood viscosity, EPT, CV, hs-CRP, IL-6. (P\u003c0.05, P\u003c0.01), and linear regression analysis showed that venous aneurysms and blood stasis were TC, LDL-C, AI, CK, LDH, cTn-1, APN, EPT, CV, hs- CRP and IL-6 (P\u003c0.01), and further linear stepwise regression analysis showed that sputum and blood congestion developed into TC, CK and it has been shown to be closely related to IL-6 (P\u003c0.01) .

  Conclusion: High-fat feeding and VD3 and isoproterenol injection can successfully establish a small model of chronic myocardial ischemia with sputum and blood congestion syndrome, lipid metabolism, hematology, myocardial enzyme metabolism and dyslipidemia. Inflammation can act as a congestion of sputum and blood. These changes are the objective biochemical material basis of mutual congestion syndrome, and are the "mutual congestion and congestion, changes in blood congestion and toxins" in Chinese medicine theory.
