Objective: To establish a rat model of type 2 diabetes with coronary heart disease.
Method: A Zucker diabetic obese rat (Zuckerdiabeticfatty, ZDF) model of type 2 diabetes and coronary heart disease was prepared by feeding a high-fat diet and continuously injecting isoproterenol hydrochloride. it is. The model was evaluated by serum creatine kinase (CK), serum creatine kinase and other enzymes (CK-MB), ECG ST and cardiac tissue morphology.
Results: The expression of CK in the non-DM model group and the DM-CHD group gradually increased with the increase in dose, and the expression of CK-MB first increased and then decreased, forming rat myocardial injury. It shows. The deviation of the ST segment of Read II of ECG in model rats indicates that myocardial injury is formed. The results of myocardial pathology section of the rats in the injection model group showed that the cardiomyocytes of the blank control group were normal; after 5 days of continuous ISO injection, the area of myocardial necrosis in ZDF rats reached 1/2, 4.4, and 1/2 of the myocardial cross section
Conclusion: CK, CK-MB content changes, ECG ST wave changes and myocardial pathological sections indicate that the model is successful. ISO may cause some myocardial damage. Continuous injection of ISO can be used to establish coronary heart disease models. In this article, we have established a simple method to establish a rat model of diabetes and coronary heart disease through a high-fat diet and continuous injection of ISO.
