OBJECTIVE: To detect the effect of low-concentration levofloxacin on Pseudomonas pneumoniae pneumonia in mice, and to explore the method of resistance to levofloxacin. Methods: PA-infected mice were used to induce a pneumonia model. The mice in each experiment were based on different bacterial concentrations of body weight, number of infections, administration methods, the law of changes in weight of mice, and mouse lung index as the endpoint for randomization. Analyze the therapeutic effect of levofloxacin and study the feasibility of establishing a model.
Result: Low concentration of levofloxacin is effective. PA (1×109 cfu/mL) mice were infected with a single infection. The lung index of model group 1 to model group 2 was significantly different from that of normal group (P\u003c0.01); levofloxacin group 2 and model group had two There are no significant differences between the participants. Low-concentration levofloxacin was infected with PA (1 x 1010 cfu/mL) in mice. Compared with the normal group, there are significant differences between model group 1 and model group 2 (P\u003c0.01). Compare Levofloxacin Group 3 with Model Group 2. Are the results significantly different? Levofloxacin 1-2 group can significantly reduce the lung index of the model group. In the experiment of PA repeatedly infected mice, is there a significant difference between model group 1 and model group 2 compared with the normal group (P\u003c 0.01)? There was no significant difference between levofloxacin group 2 and model group 2. In each experiment, the weight of the mice on the second day after infection was lower than that of the normal group. Is there any significant difference in the results?
Conclusion: Under the experimental conditions, will low-concentration levofloxacin infect mice with PA (1 x 1010 cfu/mL) once? Low concentrations of levofloxacin can be repeatedly infected with PA. Can animal models simulate a certain degree of clinical resistance?
