Objective: To establish a rat model of vaginitis and provide a new model for the development and evaluation of therapeutic agents for vaginitis.
Method: Isolate and purify clinical vaginal samples of Gardnerella vaginalis, Streptococcus albicans and Group Streptococcus patients. Gardnerellavaginalis, Streptococcus albicans and Streptococcus type B strains. In the morning, 26 female rats were pretreated with estradiol benzoate (20 mg/kg) and streptomycin (10 μg each), and the mixed strains were inoculated into the vagina in the afternoon, and 10 rats Female rats were given benzoic acid. Give a subcutaneous injection. Estradiol (20 mg/kg) was taken as the normal group, once a day for 3 consecutive days. Before grouping, collect vaginal secretions and perform vaginal irrigation, and observe mixed vaginal bacterial infections in rats. According to body weight and bacterial load (54 mg per composite metronidazole vaginal suppository, 5 consecutive days a day, every day), 20 successfully infected rats were randomly divided into model group and drug verification group. Raise 10 animals. The normal group and the model group were injected with the same amount of 0.9% sodium chloride via vagina. After the last dose, collect vaginal lavage to detect strain load, and collect vaginal secretions for vaginal smear, cleanliness and pH check. Take vaginal tissue and observe its histopathological changes by HE staining.
Result: The vaginal wall of rats in the model group became thicker, and the stratified squamous epithelial cells showed degeneration, necrosis, and depression and edema of the lamina propria. Neutrophils are the main cells, lymphocytes and single spheres infiltrate inflammatory cells, spreading spotted bleeding. The proliferation of fibroblasts in the papillary area is obvious, and the reticular fibers are slightly proliferated. The mixed bacterial count, pH value and cleanliness score of the vaginal lavage fluid of the model group were significantly higher than those of the normal group (P\u003c0.01), and the negative conversion rate was significantly reduced (P\u003c0.01). Metronidazole vaginal suppository group can significantly reduce the number of mixed bacteria in vaginal lavage fluid, pH value and cleanliness score, improve vaginal conversion rate, and significantly improve the degree of vaginal tissue lesions.
Conclusion: After pretreatment of rats with estrogen and streptomycin sulfate, mixed bacterial inoculation can establish a rat mixed vaginitis model.
