Objective: Combining high-resolution MRI and micro-CT technology to establish a rabbit femoral head hormone-induced osteonecrosis (SIONFH) model, and evaluate the repair effect of Taohong Siwu Decoction (THSWD) on SIONFH microscopic bone structure damage.
Methods: 25 New Zealand white rabbits were randomly divided into three groups: 5 in normal group, 10 in model group, Chinese herbal medicine group (taohong) combined with endotoxin (LPS) and methylprednisolone (MPS) 10) Method A hormone-induced femoral head necrosis model was established in the Siwutang group. The traditional Chinese medicine group was given Taohongtang Decoction 0.3 g/kg in the stomach, and the normal group and the model group were given the same amount of ultrapure water. In the 8th week of the experiment, the femoral head was examined by high-resolution MRI using 3.0T high-resolution magnetic resonance imaging equipment, and the femoral head was pathologically examined by hematoxylin-eosin staining and microCT examination.
Result: 1) A low signal band was seen in the femoral head of the high-resolution MRI model group. The shape of the femoral head in the traditional herbal medicine group tended to be normal, with low density. The area near the epi of the femoral skull and most of the gray values in the bone tissue are normal. Compared with the normal group, the ROE value of the former structural parameter increased, while the ROE/NR ratio of the model group increased. Compared with the model group, the ROE value of the Chinese herbal medicine group decreased, and the ROE/NR ratio decreased. The difference is significant (P\u003c0.05). 2) Hematoxylin-eosin staining showed trabecular fracture, bone cell nuclear contraction, trabecular hole, bone marrow cell necrosis in the model group, the difference was statistically significant (P\u003c0.05). The bone formation in the group is still active and the trabecular shape is better. 3) In micro CT, BV/TV, the Conn.D., SMI, Tb.N, Tb.Th of the model group are lower, and the BS/BV and Tb.Sp values are higher than those of the normal group. In the traditional herbal medicine group, BS/BV and Tb.Sp were significantly lower than the model group, Conn.D., SMI and Tb.N were all increased, and the difference was statistically significant (P\u003c0.05). OI 3D reconstruction showed that the model group was trabecular bone disease. You will see common fractures with few bones. The bone quality of the Chinese herbal medicine group was relatively improved, and the trabecula was partially repaired.
Conclusion: Taohong Siwu Decoction can promote the microstructure repair of the rabbit SION FH model and the improvement of the biomechanical environment.
