Objective: To investigate the course of acute kidney injury (AKI) in rabbits induced by a single intravenous injection of cisplatin.
Method: 40 male New Zealand rabbits were randomly divided into 5 groups with 8 rabbits in each group. A single injection of cisplatin (5 mg/kg) through the ear-marginal vein was used to make an acute kidney injury model. According to the observation time point, it was divided into 6 h group (T6), 12 h group (T12), and 24 h group (T24). ), 36 h group (T36) and 48 h group (T48), the rabbit venous blood urea nitrogen (BUN) and blood creatinine (SCr) values were measured at the corresponding time points before and after injection, respectively. SPSS 22.0 statistical software was used for statistical analysis of the data, and P<0.05 indicated that the difference was statistically significant. At each experimental node, the experimental rabbits were sacrificed, and kidney tissue specimens of each group of rabbits were obtained, and routine paraffin sections were observed under HE staining light microscope.
Results: Compared with before injection, the BUN and SCr values of each group were significantly increased after cisplatin injection. After 24 hours of modeling, all experimental rabbits in each group reached the acute kidney injury standard, and the damage was progressively worse. Observed under HE stained light microscope, the changes in T48 group were more obvious.
Conclusion: A single intravenous injection of cisplatin can cause acute renal damage in rabbits, which is progressively worse within 48 hours.
