Objective: To understand the effect of Gehua Jiejiu Dizhi Decoction on liver fat deposition in alcoholic fatty liver (AFLD) mice, and to observe its effect on liver tissue pregnane receptor (PXR) and its regulatory target genes CYP3A11 and CYP3A25 proteins.
Method: 29 male C57BL/6J mice were randomly divided into normal group (5), model group (8), high-dose intervention group (8) and low-dose intervention group (8). . The AFLD mouse model was created by the National Institute of Alcohol Abuse and Alcoholism (NIAAA) method of the National Institutes of Health, and high and low doses of Pueraria lobata Jieji Dizhi Decoction were taken orally for 9 consecutive days. Enzyme-linked immunosorbent assay was used to detect serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels, and histopathological oil red O staining was used to detect liver fat deposits, fluorescence quantitative PCR and The immunohistochemical method detects the expression of PXR, CYP3A11, CYP3A25 mRNA and PXR protein in real time.
Result: Compared with the model group, the liver fat deposition in the intervention group was significantly reduced in a dose-dependent manner, and the expression levels of PXR and CYP3A25 in the model group and the normal group were significantly increased (P). (U003c0.01); The serum ALT level of mice in the model group was significantly increased (P\u003c0.01), and the other groups were the same; there was no significant difference in the transcription level of CYP3A11 mRNA in each group (P≥.) 0.05) .
Conclusion: Homemade Gehua Jiejiu Dizhi Decoction has a significant therapeutic effect on experimental mice with AFLD. This may be related to promoting the expression of PXR and its target gene CYP3A25.
