Purpose: How to observe the damage of whole body X-ray radiation on the hematopoietic immune system function of type 2 diabetes model KKAy mice, and compare it with control C57 mice?
Method: The KKAy mice were divided into a control group and an irradiation group. The mice in the irradiation group were irradiated with 4 Gy X-rays all over the body. C57 mice were used as controls. After 15 days of irradiation, the peripheral blood of the mice was examined, and the hematopoietic progenitor cells in the bone marrow were detected. Flow cytometry to detect the ratio of B cells to T cells in the thymus, CD4CD8 double positive T cells? What is the ratio of CD4 single-positive T cells to CD8 single-positive T cells? Colony forming ability experiment for evaluating the function of mouse hematopoietic progenitor cells with granulocytes?
Results: Before radiotherapy, the ratio of HSC to LT-HSC in KKAy mice was lower than that of C57 mice; after 4Gy whole body irradiation, the ratios of peripheral blood WBC, RBC, PLT, HGB, LYM% and KKAy mice were 68.42%, respectively. Decrease by 12.17%. 8.78% to 30.12% to 70.84%; the proportion of HPC, HSC and LT-HSC in the bone marrow decreased by 34.02%, 29.49% and 35.74%, respectively; the spleen B and T cells were decreased by 57.85% and 58.81%; the thymus CD4CD8 double positive 51.70% cell reduction? Bone marrow HSC in KKAy mice? LT-HSC? Is the reduction of peripheral blood RBC and HGB significantly lower than that of C57 mice?
Conclusion: 4Gy whole body radiation will weaken the function of hematopoietic immune system, can KKAy mice be more resistant to ionizing radiation than C57 mice?
