How to prepare animal models of ischemic anemia?

Iron deficiency anemia (Ironemia anemia, IDA) refers to iron deficiency caused by various reasons and anemia caused by a decrease in red blood cells. IDA is the most common anemia, and its incidence is significantly higher in infants and women of childbearing age in economically underdeveloped areas.

[Modeling mechanism] Feeding a low-iron diet can cause iron deficiency, and regular addition of a small amount of bleeding can lead to iron deficiency anemia in animals.

[Modeling method] Choose 4-week-old Sprague-Dawley rats, male or female, weight (70±10) g, HGB≥130 g/L. Low-iron diets usually use low-iron diets recommended by the Official Association of Analytical Chemists to formulate animal feed. When the EDTA immersion method is used to remove iron from the diet, the iron in the diet is the key to the formation of an iron deficiency anemia model in Sprague-Dawley rats. After feeding animals for 2 weeks, iron deficiency anemia may occur. Supplementing a small amount of blood twice a week and supplementing 1-2 ml of tail vein blood can accelerate the formation of anemia. The bleeding continued for about 8 weeks. When the hemoglobin content of the iron-deficiency anemia group was less than 60 g/L, the bleeding stopped. After 2 weeks of observation, the hemoglobin content continued to decrease, and the animals were sacrificed at 10 weeks. Regularly observe hemoglobin content, red blood cell count and hematocrit before and after modeling to measure indicators such as serum iron, serum ferritin, transferrin saturation and total iron binding capacity. [Model Features] Iron deficiency anemia may appear 2 weeks after modeling, and severe iron deficiency anemia may appear 8 weeks after modeling.

Model indicators: ①HGB≤100g/L; ②Blood image: The number of red blood cells is less than that of normal red blood cells, the size is different, the central staining area is enlarged, the average number of red blood cells decreases, and the average red blood cell hemoglobin concentration decreases. Do it. ③ Decrease in serum iron, usually 60μmol/L

[Model Evaluation and Application] The model method is simple and the indicators are clear. After modeling, hemoglobin content decreased, red blood cell count decreased, serum iron decreased, and total iron binding capacity increased. It is usually used to study the pharmacodynamics of the treatment of iron deficiency anemia and the etiology of iron deficiency anemia.
