Objective: To explore the therapeutic effect and mechanism of Qingfei Touxue Decoction on Mycoplasma Pneumonia (MPP) mice.
Method: 40 male SPF BALB/c mice were randomly divided into control group, model group, Qingfei Touxue Decoction low-dose group, medium-dose group and high-dose group, each with 8 mice. A mouse model of Mycoplasma pneumoniae pneumonia was established by nasal injection of Mycoplasma pneumoniae (MP). The mice in the model group were given physiological saline, and the mice with low, medium and high doses of Qingfei Touxue Decoction were divided into 200 mice. The daily dosage of Qingfei Touxue Decoction suspension is 800 mg/kg body weight. After 2 weeks of treatment, the mouse lung tissue index was calculated. HE staining is used to detect pathological changes in lung tissue. TUNEL is used to detect apoptosis in mouse lungs. ELISA was used to detect IL-1β and TNF-α. The content of IL-6 and IL-18 in the alveolar lavage fluid of each mouse; qPCR to detect the expression of NLRP3, MyD88 and NF-κB mRNA in the lung tissue; Western blotting to detect the NLRP3, MyD88 and p-NF-κB protein in the lung Expression organization. Results: Qingfei Touxue Decoction can significantly reduce the lung index of MPP mice, reduce lung pathological damage and cell apoptosis rate (P\u003c0.05); Qingfei Touxue Decoction can improve the effect of alveolar lavage fluid, and also Can significantly reduce the content of IL-1β and TNF in α and IL. IL-6,, IL-18 levels (P\u003c0.05); and NLRP3, MyD88 and NF-κB mRNA and protein expression decreased (P\u003c0.05).
Conclusion: Qingfei Touxue Decoction can improve lung injury in mice with mycoplasma pneumonia, and its mechanism may be related to the inhibition of NLRP3 inflammasome and NF-κB signaling pathway.
