【Animal Modeling】-The effect of placing time of arteriovenous blood cell suspension on blood routine measurement in mice

  Objective: To observe the effect of the preservation time of venous blood cell suspension on routine blood routine of mice.

   Method: randomly select 20 adult ICR mice and C57BL/6J mice, collect quantitative venous blood, add diluent for quantitative blood cell analysis, and mix for 0 minutes and 15 minutes to prepare venous blood cell suspension. The MEK-6410C automatic blood cell analyzer is used for 2 hours, 4 hours, 0.6 hours and 8 hours for micro blood analysis to determine blood routines.

  Result: The blood routine data of ICR and C57BL/6J mice are basically the same. Compared with 0 minutes, RBC significantly decreased at 2, 4, 6, and 8 hours (P0.05).

   Conclusion: When using microblood measurement to determine the blood routine of mice, it should be completed as soon as possible after making a venous blood cell suspension. The total number of red blood cells should be determined within 1 hour. The measurement of HCT and MCV indicators related to RBC should not exceed 30 minutes. PLT measurement should be completed within 2 hours. The PCT and MPV related to PLT are determined within 1 hour. The total number of WBC and HGB content should be relatively stable.
